Yes, you should totally wet your toothbrush before applying toothpaste. 
I even go as far as wetting it after applying toothpaste.  I once this terrifying video of a wall gecko licking a toothbrush and I just can’t get that out of my head. I even use a toothbrush cover too😭. I’d rather be safe than share a toothbrush with a wall gecko imo
