Depression often involves negative thought patterns and abnormal brain functioning. Psilocybin may work by disrupting established brain networks via 5-HT2A receptor activation, promoting a temporary breakdown of rigid brain patterns and creating a wider range of possible brain states. In two clinical trials, psilocybin produced rapid and sustained antidepressant effects, linked to reduced brain network modularity and increased connectivity, especially in higher-order networks. This integration of brain networks likely underpins psilocybin’s antidepressant action. In contrast, the antidepressant escitalopram showed milder effects without brain network changes.
